转基因大豆MON89788芯片式数字PCR定量方法的建立

针对抗虫耐除草剂大豆转基因品系MON89788,从转基因植物基因组DNA的提取、核酸模板的质量和浓度控制、引物探针的筛选、PCR反应过程的建立等方面建立了一套完整的转基因大豆芯片式dPCR定量检测方法。本实验也对该方法的重复性和定量检测限进行考察。10组5%转基因品系大豆MON89788样品定量重复性RSD在1.17%-9.97%之间,均满足国际上转基因定量结果RSD小于25%的要求。用该方法对转基因含量为5%、1%、0.1%的大豆MON89788进行定量检测,其定量结果为5.20%、0.94%和0.11%,RSD分别为6.2%、3.6%和15.2%。该检测方法的定量限达到0.1%,能满足欧盟对转基因定量标识0.9%的要求。将本实验建立的方法用于转基因大豆的定量检测,能为规范我国转基因监管工作的实施提供强有力的技术支撑。     

DCMU causes conformational changes of a synthetic fragment of D1 protein: A Fourier transform infrared study

A peptide ranging from residues 229 to 240 (ENESANEGYRFG) of D1 protein was synthesized by stepwise solid-phase method. Resolution enhancement techniques were combined with band curve-fitting procedures to quantitate the FTIR spectra in the amide I' region (1700-1600 cm-1). FTIR analysis showed that DCMU induced drastic structural modification with a relative decrease of the unordered structure and turns, and a substantial increase of α-helix, which indicated that a much more compact structure was formed when DCMU was applied. The results may reflect molecular information for the protective effect of DCMU against photoinhibition.     

Development and characterization of SSR markers in lychee (Litchi chinensis)

Sixteen microsatellite loci were isolated from lychee (Litchi chinensis), all of which exhibited polymorphism (two or three alleles per locus), with levels of heterozygosity ranging from 0.021 to 0.900. These loci can help assess the genetic structure of lychee.     

Isolation and characterization of 16 tetranucleotide microsatellite loci in the red panda (Ailurus fulgens)

A total of 15 polymorphic tetranucleotide microsatellite loci in the red panda, Ailurus fulgens, were characterized in this study. Based on evaluations of 33 red pandas, the number of observed alleles for each locus ranged from seven to 17 and the expected and observed heterozygosities were 0.412–0.897 and 0.121–0.909, respectively. The mean polymorphic information content was 0.721. These markers would greatly strengthen the utilization of microsatellite tools in genetic variation studies in red panda populations.     

N-糖蛋白去糖基化酶（PNGase）的研究进展

N-糖蛋白去糖基化酶（PNGase）是一种广泛存在于真菌、植物、哺乳动物中的去糖基化酶，可以水解N-糖蛋白或 N-糖肽上天冬酰胺与寡糖链连接的化学键，并释放出完整的N-寡糖。PNGase在生物体内参与蛋白质降解、器官发育、个体生长等过程。人PNGase基因功能缺陷会导致先天性去糖基化障碍，小鼠PNGase缺陷会导致胚胎致死性，线虫PNGase缺陷使其寿命下降。本文对PNGase在不同物种的分布、蛋白质结构、酶学功能及生物学功能进行阐述，为PNGase的生理病理功能及致病机制的基础研究提供思路，为PNGase作为糖生物学工具酶或药物开发的创新应用研究奠定基础。     

美沙酮维持治疗188例海洛因成瘾者的临床疗效分析

目的：了解188例海洛因成瘾者在本门诊治疗的情况以及美沙酮口服液治疗188例海洛因成瘾者的观察分析。方法：收集本门诊由国家工作组制定的申请表及基线调查表188份,均由门诊医生实名登记和调查。结果：通过美沙维持治疗,患者和其家属均表示认可和接受,均认为是有效,治疗方法是可行的。结论：患者恢复了自信,可维持正常人的生活,67.2%的静脉注射海洛因的患者减少了注射的次数,降低了感染爱滋病的机率（见表1,P〈0.01）。但同时,如何使患者维持治疗时间的长久,需要家庭、社会多方面的支持等因素有待探讨。
